In der BBC-Miniserie dreht sich alles um das Beziehungsleben von vier Freundinnen in den Dreißigern. Das größte Problem der Damen ist, dass sie immer die falsche Person zur falschen Zeit wollen. Zum einen wäre da Katie, eine fantastische Ärztin. Sie verliebt sich in John, einen todkranken Patienten. Nach seinem Tod trifft Katie auf Sam, Johns Sohn. Er weiß von der Affäre seines Vaters und ist nun auf der Suche nach der anderen Frau. Trudi hat ihren Mann bei dem Anschlag auf das World Trade Center am 9.11. verloren und versucht nun zusammen mit ihren zwei Kindern ihr Leben in den Griff zu bekommen. Siobhan ist eine hart arbeitende Anwältin. Sie und ihr Ehemann Harry versuchen verzweifelt schwanger zu werden, was ihnen leider nicht gelingt. Als sie Dominic trifft, stellt dies ihre Ehe, die sich inzwischen dramatisch abgekühlt hat, auf eine harte Prüfung. Und für Jessica, nymphoman und beziehungsunfähig, steht der Spaß am Leben (und der Lust) an die erste Stelle. Zurzeit steckt sie mitten in einer sorglosen Affäre mit ihrem Boss und glaubt nicht daran, dass es so was wie Liebe wirklich gibt. Bis sie sich auf einer von ihr ausgerichteten lesbischen Hochzeit in Alex, eine der Bräute, verguckt. Prompt landet Jessica mit ihr im Bett … und von da an ist irgendwie alles anders …

Two years have passed and the once-unbreakable bond between best friends Katie, Trudi, Jessica and Siobhan has been shattered. As the girls gather it's clear their relationship has changed dramatically: there's tension in the room and no happy greetings. What could have happened to bring them to this?

Katie is forced to take a trip down memory lane as she visits the house she grew up in when her interfering mother, Vivienne, unexpectedly announces that she's planning to sell the property. But Katie finds more than she bargained for and unearths a disturbing secret that threatens to destroy her fond childhood memories.

Trudi is guilt-stricken, having spent the evening with Chris, and confides in Katie to seek guidance on whether she should reveal all to Richard. Katie advises her to focus on the future and forget about it, but will Trudi be able to live with her conscience?

The tension mounts as Mistresses concludes. Trudi, Jessica, Katie and Siobhan are united in grief following the death of a loved one. However, emotions run high at the funeral and, with so many questions left unanswered, grief turns to anger and a bitter row ensues, causing a huge rift between the friends.
